El anime tiene lugar en el Japón de 1868, justo durante la época de Bakumatsu (1853-1867), el fin de la era de los Shogun (Tokugawa) y precursor de la Restauración Meiji.
La historia se centra en la figura de Akizuki Yôjirô, un rônin que es el legendario Asesino Eterno o Eien no Shikaku y su búsqueda por la Cabeza del Conquistador, Hasha no Kubi, un objeto que solo aparece durante épocas de revolución y de caos. Akizuki es el portador de una espada igualmente legendaria, el Getsuruitō o el Filo de una Lágrima de Luna.
